t-Boc-N-amido-PEG6-propargyl is a click chemistry reagent containing a hexaethylene glycol (PEG6) chain, with a molecular formula of C₂₀H₃₇NO₈ and a molecular weight of 419.51. Its structure consists of an amino group protected by a tert-butyloxycarbonyl (t-Boc) group, a flexible PEG6 chain, and an alkyne terminal group. It is produced through stepwise condensation reactions: first synthesizing the Boc-protected amine compound, then reacting it with mono-methoxy PEG6, and finally introducing the propargyl group. This reagent is widely used in PROTAC molecule synthesis, where it connects E3 ubiquitin ligase ligands with target protein ligands via copper-catalyzed azide-alkyne cycloaddition (CuAAC), enabling target protein degradation.
